Varenicline displays almost 100% absorption after
oral administration and bioavailability doesn't depend of
administration time or meals. The Primary mechanism of varenicline
is clearance and renal elimination is 92% percent excreted
unchanged in the urine. Estimated half-life after oral administration
is 24 hours.
Initial studies report that using this drug more
patients were able to achieve smoking cessation after 12 weeks.
The most common adverse effect associated with varenicline
is nausea.
In summary, varenicline is a partial agonist
of the alpha-4 beta-2 nicotinic acetylcholine receptor approved
as a quit smoking medicine. It increases the percent of patients
able to achieve long-term abstinence compared to other drugs
in this category or placebo.
